登革病毒胶体金抗体检测试剂盒的研发

为建立一种检测登革病毒(Dengue virus,DENV)的胶体金抗体检测试纸条,进行抗原序列选择和同源性分析,选择特异性较高的登革病毒非结构蛋白1(Non-structural protein,NS1)检测抗体的特异性抗原,并对其特异性、灵敏性、重复性、稳定性、抗干扰性和抗破坏性进行了测试.结果显示:制备的胶体金抗体检测试剂纸条的特异性和干扰试验均符合产量质量要求,与圣路易斯脑炎病毒(St.Louis encephalitis virus,SLEV)、西尼罗河病毒(West Nile virus,WNV)、日本脑炎病毒(Japanese encephalitis virus,JEV)、黄热病病毒(Yellow fever virus,YFV)无交叉反应,试剂盒不受甘油三酯、胆红素等血清基质的影响,在 37℃保存 1个月后,仍可以达到产品的质量标准要求,同时具有较好的灵敏性.结果表明,本研究成功制备了一种快速检测登革病毒的胶体金抗体检测试纸条,具有特异性、灵敏性、重复性、抗干扰性和抗破坏性强等特点,为登革病毒检测提供了有力的工具.
Development of Colloidal Gold Antibody Detection Kit for Dengue Virus
To establish a colloidal gold antibody test strip for detecting dengue virus(DENV),we conducted antigen sequence selection and homology analysis.Subsequently,we specifically selected the specific antigen of dengue virus non structural protein 1(NS1)with high specificity for antibody detection.We then conducted tests to evaluate its specificity,sensitivity,repeatability,stability,anti-interference and anti-destruction properties.The results showed that the specificity and interference test of the prepared colloidal gold anti physical examination test agent strip met the requirements of yield and quality,and there was no cross reaction with St.Louis encephalitis virus(SLEV),West Nile virus(WNV),Japanese encephalitis virus(JEV),or yellow fever virus(YFV).Furthermore,the kit was not affected by serum matrix such as dry triglyceride and bilirubin.Even after being stored at 37℃for 1 month,it still met the product quality standards,demonstrating good sensitivity.The study successfully developed a colloidal gold antibody test strip for the rapid detection of dengue virus,characterized by specificity,sensitivity,repeatability,anti-interference and anti-destruction properties,providing a powerful tool for the detection of dengue virus.
