Preliminary Study on the Identification Technology of Mosquito Complexes Using DNA Chip
This study employed the ITS2 gene as a molecular marker for the design of hybridization probes,creating a DNA chip for the identification of eight species/subspecies within the Cules vishnui complex,Aedes albopictus subgroup,and Cx.pipiens com-plex.The findings indicate that the DNA chip,developed with the ITS2 gene as the molecular marker,is suitable for the inter-complex(subgroup)and intra-complex identification of mosquitoes,including those within the Cx.vishnui complex.The probes specific to the Ae.albopictus subgroup,Cx.pipiens complex,Cx.vishnui complex,Cx.tritaeniorhynchus,Cx.pseudovishnui,Cx.quinquefasciatus,and Cx.pipiens displayed high specificity and selectivity.Field assessments have confirmed the applicability of the DNA chip in iden-tifying conspecific mosquitoes at different developmental stages and from diverse regions.However,the chip exhibited non-specific hybridization for Cx.pipiens and Cx.quinquefasciatus.
