Study on Suitable Subculture Methods of Lactobacillus leishmannii
In this study,the traditional method of 15-day passage method was changed to 7-day passage method with liquid paraffin and glycerin broth preservation method to explore a suitable subculture method of Lactobacillus leishmannii.The standard curve and the relative standard deviation(RSD)of these two methods were analyzed through long-term experimental verification.The results showed that these two methods standard curve linear relationship r2 was>0.99,and the RSD were 2.5%-3.7%and 2.9%-7.2%,respectively.This study proved that Lactobacillus leishmannii saved by the 7-day passage method with liquid paraffin and the glycerin broth preservation method had stable activity and short activation period,which can meet the requirement of vitamin B12 detection.
