CRISPR-Cas技术是一种基于基因编辑原理的核酸检测技术,具有高特异性、高灵敏度、快速便捷、适用性广泛等优点,在核酸检测领域拥有广阔的应用前景.近年来,该技术在核酸检测中的应用取得了快速发展,已用于检测病毒、细菌、真菌、基因突变等多种核酸靶标,并展现出优于传统方法的性能.本文综述了CRISPR-Cas技术在核酸检测中的应用进展,涵盖了靶向DNA、RNA和DNA/RNA的检测方法,并对其优势和局限性进行了总结.在应用方面,CRISPR-Cas技术具有快速、便捷、灵敏的特点,能够满足口岸检验检疫快速筛查、现场检测等需求,有望成为口岸生物安全防控的新利器.
CRISPR-Cas technology,a powerful nucleic acid detection technology based on the principles of gene editing,has gained broad application prospect in the field of nucleic acid detedction due to its distinctive advantages of high specificity,high sensitivity,fast and convenient,and wide applicability.In recent years,the application of this technology in nucleic acid detection has made rapid development,and has been used to detect viruses,bacteria,fungi,gene mutations and other nucleic acid targets,and has shown better performance than traditional methods.This article overviews the application of CRISPR-Cas technology in nucleic acid detection,encompassing detection methods for targeting DNA,RNA,and DNA/RNA.Furthermore,a detailed discussion on the advantages and limitations of CRISPR-Cas technology is presented.Notably,in terms of application,the rapid,convenient,and sensitive features of CRISPR-Cas technology render it a promising tool for rapid screening and on-site detection in port inspection and quarantine,potentially serving as a new weapon in the fight against biological threats at ports.
