Comparative Analysis of the Results of Plate Counting Method and Test Plate Method for the Total Colony Count of Various Food
This paper compares the differences in the results of plate counting method,gel-based colony counting test piece and non-woven colony counting test piece for detecting colony counts in different types of samples.The colony morphology of standard strains of Escherichia coli,Staphylococcus aureus and Bacillus subtilis were compared in three types of media,namely,plate count agar,gel-type colony count test piece and non-woven colony count test piece,respectively.The colony counts of Y628 and A530 food samples were determined by the three methods,and data were analyzed to ensure the accuracy of the test results.The results show that the colony identification of gel colonies is better than that of agar and non-woven colonies,and the results are easier to read and count in different samples.The results obtained using the total number of gel-type colonies and the total number of non-woven colonies are not significantly different from the results obtained by plate count of agar.There is no significant difference between the traditional plate counting method and different types of colony test tablets on the market to determine the total number of colonies of samples Y628 and A530.Therefore,the colony test piece method can be used to determine the total number of colonies in the food mentioned above.
