病毒是一类细胞内寄生的微生物,由其引发的感染性疾病对人类健康、民生、经济等造成极大威胁.病毒侵染宿主细胞是一系列动态、精细、复杂的过程,涉及病毒与细胞多组分间的相互作用,采用传统的分子生物学手段无法实现对病毒关键生命环节的精准解析.单病毒示踪则为单个或多个病毒生命周期的原位、实时可视化研究,及侵染机制的揭示提供了新的途径.本文从单病毒示踪的意义和方法出发,展开介绍:(ⅰ)病毒的标记材料和方法;(ⅱ)病毒成像与分析技术;(ⅲ)单病毒示踪的研究现状与趋势,并针对该领域目前存在的问题及未来发展趋势进行探讨.
Single virus tracking
Viruses are a kind of microorganisms,which cause infectious diseases and pose a great threat to human health,economy,and lifestyle.Virus infection includes a series of dynamic,delicate and complex processes,involving the interaction between virus and cell components.It is difficult to accurately analyze the vital lifecycle of virus infection by traditional molecular biological methods.Single virus tracking provides a novel way for in situ and real-time visualization of the life cycle of single or multiple viruses,as well as for revealing infection mechanisms.This paper introduced the following parts:(ⅰ)virus labeling materials and methods;(ⅱ)imaging equipment and analysis platform;(ⅲ)research progress of single virus tracking.In addition,the current problems and future development trends in this field were discussed.
