Chinese medicine formula research strategy based on compound compatibility
The efficacy of traditional Chinese medicine formulas has been validated during its long-term clinical practice. However, controlling their safety, effectiveness, and quality stability is difficult owing to their complex chemical composition.Although studies on component-based Chinese medicines have promoted the development of traditional Chinese medicine, further elucidating their targets and molecular mechanisms of action is still difficult, and the scientific connotation of the formula compatibility theory remains unclear. Thus,we propose a research strategy for Chinese medicine formulas based on active compound compatibility,i.e.,compound-based Chinese medicine formula(CCMF).The key technology challenges include the following:(ⅰ)analysis of chemical composition absorbed into the blood or tissues,(ⅱ)construction of a three-dimensional screening model,(ⅲ)research on functional targets, and(iv)data integration and analysis.CCMF possesses not only advantages such as synergistic effects and toxicity reduction for traditional prescriptions with compatibility features but also a clear composition similar to modern medicines.Furthermore, the scientific connotation of the formula compatibility theory can be clearly elucidated using multidisciplinary technology to verify targets and synergistic molecular mechanisms of CCMF.This is an inevitable direction of the development of research on traditional Chinese medicine prescriptions in the future. The research strategy proposed in this study can help address major research challenges, such as the scientific interpretation of the original theories of traditional Chinese medicine from a methodological perspective.
