摘要
阿尔茨海默病(AD)是一种进行性神经退行性疾病,其主要病理特征包括β淀粉样蛋白(Aβ)聚集、过度磷酸化tau蛋白和神经炎症,等.半胱氨酸蛋白酶募集域的凋亡相关斑点样蛋白(ASC)是炎症小体的重要接头蛋白.研究发现ASC与AD的发生发展密切相关,ASC可通过激活炎症反应,加速Aβ聚集,促进神经元凋亡,加重AD病理发展.此外,ASC介导tau蛋白的炎症反应通路参与AD的病理过程.文中将从ASC的结构和功能出发,详细阐述ASC在AD发病机制中与Aβ和tau蛋白的作用,旨在为AD的治疗提供新的靶点.
Abstract
Alzheimer's disease(AD)is a progressive neurodegenerative disorder whose main pathological features include β-amyloid protein aggregation,hyperphosphorylated tau protein,and neuroinflammation.The apoptosis-associated speck-like protein containing a caspase recruitment domain(ASC),is an important junction protein of inflammasomes.ASC can participate in the occurrence and development of AD by activating inflammatory responses,accelerating Aβ aggregation,and promoting neuronal apoptosis.In addition,ASC may influence the pathological process by mediating the inflammatory response pathway of tau protein.The mechanism of ASCs mediating Aβ and tau in the pathogenesis of AD is detailed from the structure and function of ASC,in order to provide new targets for the treatment of AD.
