The effect of Zhu Lian's excitation type Ⅰ acupuncture at different periods on the ROCK and PI3K/AKT pathways in diabetes neurogenic bladder rats
Objective To observe the effect of Zhu Lian's excitation type Ⅰ acupuncture at different periods on Rho associat-ed protein kinase(ROCK)and phosphatidylinositol-3-hydroxykinase(PI3K)/protein kinase B(AKT)pathways in rats with diabetes neurogenic bladder(DNB),and to explore the mechanism of this acupuncture in treating DNB.Methods Fifty SD rats were randomly divided into a control group,a model group,an acupuncture prevention group,an acupuncture treatment group,and an acupuncture prevention+treatment group,with 10 rats in each group.The DNB model of rats was established by intraperitoneal injection of strepto-zotocin.The"Zhongji""Sanyinjiao""Lieque"and"Taichong"acupoints of rats was selected by Zhu Lian's acupuncture.The acu-puncture prevention group was bound with acupuncture every day from 6 to 9 w,the acupuncture treatment group was bound with acu-puncture every day from 10 to 13 w,and the acupuncture prevention+treatment group was bound with acupuncture every day from 6 to 13 w.If there was no acupuncture intervention in each group,routine binding was performed every day.The maximum bladder capaci-ty,residual urine volume,and bladder wet weight of each group were compared;bladder tissue morphology was observed by hematoxy-lin-eosin staining;Western blot was used to detect the expressions of PI3K and AKT protein;enzyme linked immunosorbent assay(ELISA)was used to measure the expressions of ROCK1,ROCK2,P-PI3K,and P-AKT protein;Real time fluorescence quantitative polymerase chain reaction was used to detect the expressions of ROCK1,ROCK2,PI3K,and AKT mRNA.Results Compared with the control group,maximum bladder capacity,residual urine volume,and bladder wet weight were increased,expressions of ROCK protein and mRNA were increased,expressions of PI3K/AKT protein and mRNA were decreased in the model group,acupuncture pre-vention group,acupuncture treatment group,and acupuncture prevention+treatment group,the difference was statistically significant(P<0.05).Compared with the model group,maximum bladder capacity,residual urine volume,and bladder wet weight were de-creased,expressions of ROCK protein and mRNA were decreased,expressions of PI3K/AKT protein and mRNA were increased in the acupuncture prevention group,acupuncture treatment group,and acupuncture prevention+treatment group,the difference was statisti-cally significant(P<0.05).The maximum bladder capacity and residual urine volume in the acupuncture treatment group,acupuncture prevention group,and acupuncture prevention+treatment group had no significant difference(P>0.05),the wet weight of the bladder was decreased sequentially,the protein and mRNA expression levels of ROCK protein and mRNA were decreased sequentially,the pro-tein and mRNA expressions of PI3K/AKT were increased sequentially,the difference was statistically significant(P<0.05).Conclu-sions Zhu Lian's excitation type Ⅰ acupuncture method is beneficial for improving bladder function in rats,and its mechanism may be related to its downregulation of ROCK expression in the bladder tissue of DNB rats and upregulation of PI3K/AKT pathway expres-sion.The effect of preventive intervention is better than that of post attack intervention,and the combination of prevention and treat-ment is better than simple preventive intervention or post attack intervention.
