Chemosensitization of HCT116 colon cancer cells by α-hederin in combi-nation with oxaliplatin
Objective To investigate the chemosensitizing effect of combining α-hederin(HED)with oxaliplatin(OXA)on HCT116 colorectal cancer(CRC)cells.Methods The MTT assay was used to detect the inhibitory effects of α-HED and OXA,both individually and in combination,on the proliferation of HCT116 cells.Synergistic interactions and optimal dosing regimens were ana-lyzed using CompuSyn software.Plate colony formation assays,flow cytometry,and Western blot analysis were conducted to evaluate the combined effects of α-HED and OXA on colony formation and apoptosis in HCT116 cells.For in vivo experiments,a subcutaneous xenograft model was established by implanting HCT116 cells into nude mice,which were divided into Control,α-HED,L-OXA,N-OXA,and α-HED+L-OXA groups.The effects of α-HED and OXA,individually and in combination,on tumor growth in the CRC model were subsequently observed.Results The inhibitory effects of 25 μmol/L OXA on both the proliferation and colony formation rate of HCT116 cells were significantly enhanced by 6 μmol/L α-HED,apoptosis was significantly promoted by 6 μmol/L α-HED(P<0.05).Both α-HED and OXA alone effectively inhibited the growth and proliferation of CRC xenografts in nude mice,the antitumor effect was significantly more pronounced when the two agents were administered in combination(P<0.05).α-HED and OXA were ad-ministered in combination which were significantly downregulated the expressions of Ki67 and CD31,with a more significant reduction observed in comparison to the α-HED and low-dose OXA groups(P<0.05).Conclusions The combination of α-HED and OXA could exhibite chemosensitization effects on HCT116 CRC cells both in vitro and in vivo.
