Effects of herb pair of Rhei Radix et Rhizoma and Salviae Miltiorrhizae Radix et Rhizoma with different medicinal guides in pulmonary fibrosis
Objective To compare the effect difference of herb pair of Rhei Radix et Rhizoma and Salviae Miltiorrhizae Radix et Rhizoma with different medicinal guides in pulmonary fibrosis of rats,and to explore the synergistic effect of medicinal guides.Meth-ods Sixty male SD rats were randomly divided into sham operation group,model group,Salvia Miltiorrhiza Radix herb pair group(herb pair for short),herb pair+Platycodonis Radix group,herb pair+Euodiae Fructus group and herb pair+Asari Radix et Rhizom group.Except sham operation group,the pulmonary fibrosis model was established by slowly injecting 5 mg/kg bleomycin into the tra-chea.From the second day of the model,each group was given corresponding drugs by intragastric administration for 2 w.Blood and lungs were taken for blood gas analysis after the last administration.The histopathology of lung tissue was observed by hematoxylin-eo-sin(HE)and Masson staining.The contents of malondialdehyde(MDA),superoxide dismutase(SOD),hydroxyproline(HYP)and reduced glutathione(GSH)in lung tissue were determined by enzyme linked immunosorbent assay(ELISA).The protein expressions of transforming growth factor(TGF)-β1,SMAD homologue(Smad)3,phosphorylated(p)-Smad3 and α-smooth muscle actin(SMA)in lung tissue were analyzed by Western blot.Results Compared with the model group,the pathology of lung tissue in each experi-mental group was significantly improved,the lung coefficient,the contents of MDA and HYP in lung tissue were significantly de-creased,the levels of partial pressure of arterial oxygen(PaO2)in arterial blood,SOD and GSH in lung tissue were increased signifi-cantly,and the expressions of TGF-β1,p-Smad3 and α-SMA in lung tissue were significantly decreased(P<0.05).Compared with the herb pair group,the lung coefficient and the contents of MDA and HYP in the lung tissue were significantly decreased,PaO,in arterial blood and the level of GSH in the lung tissue were significantly increased,and the expressions of TGF-β1,p-Smad3 and α-SMA pro-tein in the lung tissue were significantly decreased in the herb pair+Platycodonis Radix group(P<0.05).Conclusions The herb pair of Rhei Radix et Rhizoma and Salviae Miltiorrhizae Radix et Rhizoma could improve pulmonary fibrosis in rats,and its mechanism may be related to the inhibition of TGF-β1/Smad3 signal pathway.Platycodonis Radix could enhance the anti-pulmonary fibrosis effect of rhubarb-Salvia miltiorrhiza.
