The effect and molecular mechanism of IL-17A derived from lung cancer A549 cells on osteoclast differentiation
Objective To study the effect of interleukin(IL)-17A derived from lung cancer A549 cells on the differentiation of osteoclast and its molecular mechanism.Methods Lentivirus interference technology was used to construct A549 cells with low expres-sion of IL-17A(A549-si).A549 cells were co-cultured with osteoclast precursor cell to observe the effect of A549-si on osteoclast dif-ferentiation.The effect of A549-si on the expression of osteoclast differentiation related genes and IL-8 mRNA was detected by fluores-cent quantitative polymerase chain reaction(qPCR).The effect of A549-si on IL-8 protein expression was detected by enzyme linked immunosorbent assay(ELISA).IL-8 antibody was applied to the co-culture model of A549 cells and osteoclast to observe whether IL-8 antibody could weaken the promoting effect of A549 cells on osteoclast differentiation.Results The differentiation of osteoclast was sig-nificantly promoted by co-culture of A549 cells with osteoclast precursor cell.The promotion of A549 cells on osteoclast differentiation and the expression of osteoclast differentiation related genes were significantly inhibited by A549-si(P<0.05).The expressions of IL-8 mRNA and protein in A549 cells were significantly inhibited by A549-si;the promoting effect of A549 cell on osteoclast differentiation was significantly reversed by IL-8 antibody(P<0.05).Conclusions IL-17A derived from A549 cells could promote the differentiation of osteoclast by promoting the expression of IL-8.
