Effects of Natural Component of Zanthoxylum Xanthoxylum(WGX-50)on Moisture Retention and Skin Barrier Function
Objective To study the moisturizing properties of the natural component of Zanthoxylum WGX-50 and its effect on skin barrier function.Methods Keratinocyte HaCaT and 16 subjects with good skin condition were studied.HaCaT cells were used to test the cytotoxicity of WGX-50,and the effects of WGX-50 on the expression levels of aquaporin 3(AQP3),Filaggrin,Claudin-1,hyaluronic acid synthetase 2(HAS2)and CD44 were analyzed.Subjects were used to test the effects of WGX-50 on skin barrier function.Results 0.5 μmol/L WGX-50 promoted HaCaT cell proliferation(P<0.05),and 1.0-100.0 μmol/L WGX-50 inhibited HaCaT cell activity,and the cell activity was lower than that of control group(P<0.05).In particular,high concentration of WGX-50(≥10 μmol/L)significantly inhibited the activity of HaCaT cells(P<0.01).0.5 μmol/L WGX-50 could effectively promote AQP3 gene expression(P<0.05),but had no significant effect on Filaggrin,Claudin-1,HAS2 and CD44 gene expression levels(P>0.05).Experiments on skin moisture content and transepidermal moisture loss rate showed that xanthoxylin WGX-50 had good moisturizing effect on skin.Conclusion Low concentration of xanthoxylin(0.5μmol/L)WGX-50 can promote the proliferation of HaCaT cells,enhance the expression of AQP3 gene in HaCaT cells,and improve the function of human skin barrier.
