Optimization of the Culture Conditions for Three Efficient Strains Used in Ramie Bio-degumming
The optimal culture conditions of three efficient strains used in ramie bio -degumming were researched. Cell suspension turbidity (optical density at 600 nm,OD600 )was measured using spec-trophotometer to investigate the optimal initial pH,inoculation amount,culture temperature and time of the activation conditions for efficient strain used in ramie bio-degumming. Using one-factor-at-a-time combined with orthogonal array design method,optimal combination of the activation conditions for the three aimed efficient strains were found. Results showed that Hn1 -1 strain achieved the highest bio-mass with inoculation amount 5% in 8 h of incubation at initial pH 6.0 and 35.5℃,Hn2 -2 strain a-chieved the highest biomass with inoculation amount 6%in 7 h of incubation at initial pH 8.0 and 30℃, and Hn6-2 strain achieved the highest biomass with inoculation amount 7%in 6 h of incubation at ini-tial pH 6.0 and 35℃in the given conditions. Therefore,it can provide scientific basis for further appli-cation in ramie degumming as for the three improved liquid biomass (as 1.5 ~2.0 folds as that of original OD600 )by optimizing the culture conditions.
