Development of KASP Marker Set for Screening and Identification of Industrial Hemp Germplasm Resources
Currently,the molecular screening and identification of industrial hemp germplasm re-sources in China mainly rely on methods such as AFLP,SSR and Resequencing,lacking in a fast,accu-rate,and intuitive molecular marker system.The development of Kompetitive Allele Specific PCR(KASP)with marker sets and its detection system has good application prospects.This study first suc-cessfully developed a specific KASP locus based on the cannabinoid B locus gene polymorphism,which can accurately distinguish different chemical types of low-THC,high-THC or intermediate types in can-nabis germplasm resources.A total of 32 high-quality core KASP markers were developed based on the whole-genome resequencing single nucleotide polymorphism(SNP)data from 95 different cannabis germ-plasm resources.Verification experiment showed that the 33 KASP marker sets and its detection system can be applied to industrial hemp assisted breeding,molecular identification of industrial hemp varieties,and fingerprint library construction,enriching the types and methods of industrial hemp molecular mark-ers.
