水族祛痹方对KOA兔超声表现、病理评分及MMP-3、Collagen Ⅱ表达的影响

摘要
目的:探讨水族祛痹方对KOA兔超声表现、病理评分及MMP-3、Collagen Ⅱ表达的影响.方法:将30只长耳大白兔随机分为正常组、模型组、水族祛痹方组,每组10只.以木瓜蛋白酶膝关节腔内注射诱导建立兔膝骨关节炎模型,给与水族祛痹方水煎剂治疗,每日2次.造模后第28天比较超声下关节病变及病理评分,免疫组化检测关节软骨MMP-3、Collagen Ⅱ的表达水平.结果:与正常组比较,模型组超声观察Ridit值及病理Mankins评分明显升高(P＜0.05),膝关节软骨MMP-3表达水平明显上升(P＜0.05),Collagen Ⅱ表达水平明显下降(P＜0.05).与模型组比较,水族祛痹方组超声观察Ridit值及病理Mankins评分均明显降低(P＜0.05),膝关节软骨MMP-3表达水平明显下降(P＜0.05),Collagen Ⅱ表达水平明显上升(P＜0.05).结论:水族祛痹方可以减轻MMP-3对软骨基质的降解,减少软骨胶原丢失,从而减轻滑膜增厚和血管增生,延缓关节退变.
Abstract
Objective To study the effect of Shui People Qubi Formula on the ultrasonic performance,the pathological evaluation,the expression level of MMP-3 and Collagen Ⅱ in KOA rabbits.Methods 30 white rabbits were randomly divided into normal group,model group,and Shui People Qubi Formula treatment group,10 rabbits in each group.Knee osteoarthritis model was established by intra-articular injection of papain.The rabbits were treated twice a day.28 days after treatment,the joint characteristic of each group was evaluated by ultrasound and pathological detection separately,and MMP-3 and Collagen Ⅱ expression level in articular cartilage were detected by immunohistochemistry.Results Compared with the normal group,the Ridit value and pathological Mankins score of the model group rabbits significantly increased(P＜0.05),MMP-3 expression in knee articular cartilage significantly increased(P＜0.05),and Collagen Ⅱ expression significantly decreased(P＜0.05).Compared with the model group,the R value and pathologi-cal Mankins score of rabbits in the Shui People Qubi Formula treatment group significantly decreased(P＜0.05),MMP-3 expression in knee articular cartilage significantly decreased(P＜0.05),and Collagen Ⅱ expression significantly increased(P＜0.05).Conclu-sion Shui People Qubi Formula can reduce the degeneration of cartilage matrix causing by MMP-3,it also can alleviate synovial prolif-eration and reduce knee joint angiogenesis,which is to delay joint degeneration.
