摘要

目的:结合单因素试验,利用Box-Behnken设计原理设计响应面法优化大果沙枣总黄酮、多酚的提取工艺.方法:在单因素试验基础上,以提取时间、液料比、乙醇浓度为黄酮考察因素,以提取时间、液料比、丙酮体积分数为多酚考察因素,采用响应面法优化提取工艺条件.结果:大果沙枣总黄酮最优提取条件是当提取时间为28.7 min,液料比为17.25∶1,乙醇浓度为57.6%时,总黄酮提取率达到最大值4.15%.大果沙枣总多酚最优提取条件是当提取时间为40.55 min,液料比为10.32∶1,丙酮体积分数为51.1%时,总多酚提取率达到最大值1.19%.结论:本方法提取大果沙枣中总黄酮、多酚的工艺合理,重现性良好,可以为大果沙枣的深度开发提供理论依据.

Abstract

Objective Combined with the single-factor test,the Box-Behnken design principle was used to design the response surface method to optimize the extraction process of total flavonoids and polyphenols from Elaeagnus Moorcroftii Wall.ex Schlecht.Methods On the basis of the single-factor test,the extraction time,liquid-material ratio and ethanol concentration were taken as the flavonoid investigation factors,and the extraction time,liquid-material ratio and acetone volume fraction were taken as the poly-phenol investigation factors,and the response surface method was used to optimize the extraction process conditions.Results The opti-mal extraction conditions of total flavonoids:when the extraction time was 28.7 min,the liquid-material ratio was 17.25∶1,and the ethanol concentration was 57.6%,the extraction rate of total flavonoids reached the maximum value of 4.15%.The optimal extraction conditions of total polyphenols:when the extraction time was 40.55 min,the liquid-material ratio was 10.32∶1,and the volume frac-tion of acetone was 51.1%,the extraction rate of total polyphenols reached the maximum value,which was 1.19%.Conclusion The extraction of total flavonoids and polyphenols from Elaeagnus Moorcroftii Wall.ex Schlecht is reasonable and reproducible,which can provide a theoretical basis for the in-depth development of Elaeagnus Moorcroftii Wall.ex Schlecht.
