Effects of Memantine combined with Donepezil in treatment of patients with Alzheimer's disease
Objective:To observe effects of Memantine combined with Donepezil in treatment of patients with Alzheimer's disease(AD).Methods:A prospective study was conducted on 82 patients with AD admitted to the affiliated hospital of Beihua university from April 2020 to April 2023.According to the random number table method,they were divided into control group and observation group,41 cases in each group.The control group was treated with Donepezil,while the observation group was treated with Memantine on the basis of that of the control group.Both groups were treated for 8 weeks.The clinical efficacy,the scores of cognitive function[mini-mental state examination(MMSE),the Alzheimer's disease assessment scale-cognitive section(ADAS-cog)]and the activities of daily living[activities of daily living(ADL)],the neurotransmitter indexes[5-hydroxytryptamine(5-HT),dopamine(DA),acetylcholine(Ach)]levels,and incidence of adverse reactions were compared between the two groups before and after the treatment.Results:The total effective rate of the observation group was 95.12%(39/41),which was higher than 78.05%(32/41)of the control group,and the difference was statistically significant(P<0.05).After the treatment,the MMSE and ADL scores of the observation group were higher than those of the control group,the ADAS-cog score was lower than that of the control group,and the differences were statistically significant(P<0.05).After the treatment,the levels of 5-HT,DA and Ach in the observation group were higher than those in the control group,and the differences were statistically significant(P<0.05).However,there was no significant difference in the incidence of adverse reactions between the two groups(P>0.05).Conclusions:Memantine combined with Donepezil in the treatment of the AD patients can improve the total effective rate,the activities of daily living scores and the neurotransmitter index levels,and improve the cognitive function scores.Moreover,it is superior to single Donepezil treatment.
