目的:探究芪麦益心颗粒对左冠状动脉前降支结扎导致急性心肌缺血大鼠心功能及心肌组织凋亡的影响,并为其临床应用提供依据.方法:将Wistar大鼠随机分成对照组、模型组、芪麦益心颗粒组、单硝酸异山梨酯组.对照组、模型组给予蒸馏水灌胃,芪麦益心颗粒组2.43 g/(kg·d)生药灌胃、单硝酸异山梨酯组1 mg/(kg·d)生药灌胃.除对照组外,其余各组大鼠均以左冠状动脉前降支结扎法造模,观察大鼠血清CPK、LDH浓度,RT-PCR检测心肌TNFα、Fas mRNA,Western Blot检测心肌Fas、Caspase3表达水平.结果:芪麦益心颗粒可以降低血CPK、LDH浓度,下调TNFα、Fas转录水平的表达,下调Fas、Caspase3蛋白水平的表达.结论:芪麦益心颗粒对急性心肌缺血大鼠心肌具有保护作用,其作用机制可能与下调死亡途径的受体和配体从而拮抗心肌组织的凋亡有关.
Objective:To investigate the effects of Qi Mai Yi Xin granule(QMG)on cardiac function and myocardial tissue apoptosis in rats with acute myocardial ischemia induced by left anterior descending coronary artery(lad)ligation,and to provide evidence for its clinical application.Methods:Wistar rats were randomly divided into control group,model group,QMG Group and Isosorbide Mononitrate group.The Rats in the control group and the model group were treated with distilled water,the rats in the QMG group were treated with herbal medicine 2.43 g KG·d and the rats in the Isosorbide Mononitrate group were treated with herbal medicine 1mg KG·d.Except the control group,the rats in the other groups were made by ligating the left anterior descending coronary artery.The concentrations of CPK and LDH in serum were observed.The expression of TNF-α and Fas mRNA in myocardium was detected by RT-PCR,and the expression of FAS and Caspase-3 in myocardium was detected by Western blot.Results:qmg could decrease the concentration of CPK and LDH,down-regulate the expression of TNF α and Fas,down-regulate the expression of FAS and caspase 3.Conclusion:QMG has a protective effect on myocardium of rats with acute myocardial ischemia,and its mechanism may be related to down-regulation of death pathway receptor and ligand to antagonize myocardial apoptosis.
