蒙药健胃十味丸对乙醇所致大鼠慢性胃炎的保护作用研究

摘要
目的:本研究利用乙醇所致的大鼠慢性胃炎动物模型,验证蒙药健胃十味丸的治疗作用,旨在为蒙药健胃十味丸的临床应用提供理论依据.方法:选择雄性SD大鼠,除空白组外,其余各组灌胃56％乙醇造模,每周2次,连续4周,模型成功后各给药组分别灌胃不同剂量的健胃十味丸和阳性药替普瑞酮.给药结束后麻醉取血、取胃组织,对胃组织进行宏观观察、HE染色及PAS染色,检测大鼠血清及胃组织中的IL-6、IL-8、TNF-α、VEGF-A及NO的表达.结果:健胃十味丸可以显著恢复胃黏膜表面上皮细胞数量,保护胃黏膜损伤,降低大鼠血清及胃组织中促炎细胞因子的表达水平.结论:健胃十味丸能够恢复胃黏膜损伤,抑制炎症水平,抑制炎-癌转化,从而治疗乙醇所致的慢性胃炎.
Abstract
Objective:In this study,the animal model of chronic gastritis induced by ethanol in rats was used to verify the therapeutic effect of Mongolian medicine Jianwei Shiwei Pills,aiming to provide a theoretical basis for the clinical application of Mongolian medicine Jianwei Shiwei Pills.Methods:Male SD rats were selected.Except for the blank group,the other groups were given 56％ethanol by gavage twice a week for four weeks.After the success of the model,each administration group was intragastrically administered with different doses of Jianwei Shiwei Pills and positive drug teprenone.After the end of the administration,the blood was taken and the gastric tissue was taken.Macroscopic observation,HE staining and PAS staining were performed on the gastric tissue to detect the expression of IL-6,IL-8,TNF-α,VEGF-A and NO in serum and gastric tissue of rats.Results:Jianwei Shiwei Pills can significantly restore the number of epithelial cells on the surface of gastric mucosa,protect gastric mucosal injury,and reduce the expression of pro-inflammatory cytokines in serum and gastric tissue of rats.Conclusion:Jianwei Shiwei Pills can restore gastric mucosal injury,inhibit the level of inflammation,inhibit inflammation-cancer transformation,and thus treat chronic gastritis caused by ethanol.
