Determination of ursolic acid and oleanolic acid in elderberry from different areas of Inner Mongolia by HPLC
Objective:To establish a HPLC method for the determination of ursolic acid and oleanolic acid in Mongolian medicine Sambucus williamsi Hance.Methods:Promosil c18 column(250mm × 4.6mm)was used,acetonitrile-methanol-0.5%ammonium acetate solution(59∶14∶27)was used as the mobile phase,isometric elution,column flow rate was 1.0mL·min-1,detection temperature 40℃,wavelength 210nm.Results:the average contents of oleanolic acid and ursolic acid were 0.3021and 0.585mg/g,respectively.There was a good linear relationship between ursolic acid and ursolic acid.The r values of ursolic acid and ursolic acid were 0.9995 in 1.048~3.143ug/ul and 0.9996 in 0.778~2.333ug/ul.The average recoveries of ursolic acid and oleanolic acid were 102.83%and 99.02and the RSD was 1.08 and 1.88 respectively.Conclusion:in summary,HPLC is a scientific and effective method for the determination of ursolic acid and oleanolic acid.The detection results have high accuracy and precision,and can be used to evaluate the quality of Mongolian medicine Sambucus williamsi Hance.
