Objective:To investigate the effect of Mongolian medicine Eleven Flavors Dingxiang Powder on airway remodeling and epithelial mesenchymal transformation(EMT)in mice with bronchial asthma induced by house dust mites.Method:Forty male BALB/c mice were randomly divided into four groups,with 10 mice in each group,namely the blank group,model group,Mongolian medicine Eleven Flavors Dingxiang Powder group,and budesonide group.A bronchial asthma model was replicated using the house dust mite nose drop method.Measure the basal enhanced expiratory interval(Penh)value of mice stimulated by methacholine for airway hyperresponsiveness(AHR)detection,observe the pathological and morphological changes of lung tissue in each group with HE staining,and detect E-cadherin and N-cadherin in mouse lung tissue with Western blot and RT-PCR methods α The expression levels of smooth muscle actin(a-SMA)protein and mRNA.Result 1.Compared with the blank group,the model group showed a significant increase in Penh values,with a statistically significant difference(P<0.01);Compared with the model group,the Penh values in the budesonide group and the Mongolian medicine Eleven Flavors and Cloves powder group decreased,with a statistically significant difference(P<0.01);Compared with the budesonide group,there was no statistically significant difference(P>0.05)between the Mongolian medicine Eleven Flavors Dingxiang Powder group and the budesonide group.2.Compared with the blank group,the pathological morphology of lung tissue in the model group showed severe abnormalities in lung tissue structure,with a large number of alveolar atrophy;Compared with the model group,the Mongolian medicine Eleven Flavored Cloves group and Budesonide group significantly reduced airway inflammation and airway mucosal thickness.3.Compared with the blank group,the detection of E-cadherin,N-cadherin,and a-SMA in the model group showed a significant decrease in the expression of E-cadherin protein and E-cadherin mRNA,while the expression of N-cadherin protein and N-cadherin mRNA increased significantly.The difference was statistically significant(P<0.01);Compared with the model group,the expression of N-cadherin protein and N-cadherin mRNA decreased,while the expression of E-cadherin protein and E-cadherin mRNA increased,while the expression of a-SMA protein and a-SMA mRNA decreased in the Mongolian Medicine Eleven Flavor Dingxiang Powder group and Budesonide group,with statistical significance(P<0.01);There was no statistically significant difference(P>0.05)between the Mongolian medicine Eleven Flavor Dingxiang Powder group and the Budesonide group.Conclusion:Mongolian medicine Eleven Flavors Dingxiang Powder can reduce airway hyperresponsiveness and improve airway remodeling in asthma mice,and its mechanism of action may be related to the regulation of EMT.
