To screen the rapid propagation scheme of passion fruit by tissue culture and provide technical sup-port for the breeding of passion fruit tissue culture seedlings,the effects of different sterilization times on the survival rate of explants and different medium compositions on bud induction,subculture proliferation,rooting and transplanting were studied using stem segments of yellow passion fruit as explants.The results showed that treatment of 0.1%HgCl2 for 6 minutes had the best sterilization effect on passion fruit stem explants,with a survival rate of 66.67%.The culture medium of MS+0.3 mg·L-1 6-benzylaminopurine(6-BA)+0.1 mg·L-1 indole-3-butyric acid(IBA)+300 mg·L-1 ascorbic acid was the optimal medium for bud in-duction with the bud induction rate of 79.14%.The best subculture proliferation medium was MS+0.2 mg·L-1 6-BA+0.05 mg·L-1 IBA+20 mg·L-1 glutamic acid,with a proliferation rate of 3.21.The MS+0.5 mg·L-1 IBA was the optimal rooting medium with a 100%rooting rate,and was conducive to the survival of the transplanted tissue culture seedlings.
