为筛选黄百香果组培快繁方案,为百香果组培苗繁育提供技术支撑,以黄百香果茎段为外植体,研究不同灭菌时间对外植体成活率的影响,不同培养基配方对初代芽诱导、继代增殖、生根移栽的影响.结果表明,0.1%HgCl2处理6 min对百香果茎段外植体的灭菌效果最佳,外植体成活率为66.67%.MS+6-苄氨基嘌呤(6-BA)0.3 mg/L+吲哚丁酸(IBA)0.1 mg/L+抗坏血酸300 mg/L为芽诱导最佳培养基,芽诱导率79.14%.MS+6-BA 0.2 mg/L+IBA 0.05 mg/L+谷氨酸20 mg/L为继代增殖最佳培养基,增殖倍数3.21.MS+IBA 0.5 mg/L生根培养基的组培苗生根率100%,且有利于组培生根苗移栽成活.
Research on Tissue Culture and Rapid Propagation of Stem Segments of Yellow Passion Fruit
To screen the rapid propagation scheme of passion fruit by tissue culture and provide technical sup-port for the breeding of passion fruit tissue culture seedlings,the effects of different sterilization times on the survival rate of explants and different medium compositions on bud induction,subculture proliferation,rooting and transplanting were studied using stem segments of yellow passion fruit as explants.The results showed that treatment of 0.1%HgCl2 for 6 minutes had the best sterilization effect on passion fruit stem explants,with a survival rate of 66.67%.The culture medium of MS+0.3 mg·L-1 6-benzylaminopurine(6-BA)+0.1 mg·L-1 indole-3-butyric acid(IBA)+300 mg·L-1 ascorbic acid was the optimal medium for bud in-duction with the bud induction rate of 79.14%.The best subculture proliferation medium was MS+0.2 mg·L-1 6-BA+0.05 mg·L-1 IBA+20 mg·L-1 glutamic acid,with a proliferation rate of 3.21.The MS+0.5 mg·L-1 IBA was the optimal rooting medium with a 100%rooting rate,and was conducive to the survival of the transplanted tissue culture seedlings.
