Determination of genome size of strawberry with different ploidy based on flow cytometry
The young leaves of strawberry with different ploidy were used as materials.Rice variety Nippon-bare(genome size of 430 Mb),tomato(genome size of 900 Mb),and maize B73(genome size of 2.3 Gb)were selected as internal references according to the estimated genome size of ploidy level of the testing samples.The genome size of the samples was determined by flow cytometry.The results showed that for diploid re-sources,the genome size of Fragaria vesca as(233±0.78)Mb,and the F.viridis was(234±0.82)Mb.For tetraploid resources,the genome size of F.moupinensis was(492±2.12)Mb,F.corymbosa(male plant)was(488±7.07)Mb,and the F.corymbosa(female plant)was(491±4.95)Mb,with the genome si-zes of the male and female plants of the F.corymbosa being determined for the first time.For hexaploid re-sources,the genome size of F.moschata was(644±1.73)Mb.For octoploid resources,the cultivated variety F.ananassa Akihime had a genome size of(793±7.78)Mb,and Monterey was(761±0.00)Mb.For deca-ploid resources,the wild resources of F.iturupensis had a genome size of(938±6.36)Mb,while the culti-vated variety Tao Xun was(1,007±6.36)Mb.
