Cloning and analysis of homologous sequences of NBS-LRR disease resistant genes of Passiflora foetida L.
Passiflora foetida L.is a closely related wild species of the cultivated species of genus Passiflora,with strong stress resistance and wide adaptability.It is a potential gene resource pool for disease resistance breeding of Passiflora.Degenerate primers were designed based on the conserved domain of the known plant disease resistance gene NBS-LRR to clone NBS-LRR class disease resistance gene homologous sequences(RGAs)from the P.foetida L.genome using P.foetida L.as the test material,and perform cluster analy-sis.The results showed that there were two types of P.foetida L.NBS-LRR disease resistance gene se-quences,TIR and non TIR.Among them,there were 5 TIR-NBS-LRR types of sequences,numbered from LRGA-1 to LRGA-5,and 7 non TIR-NBS-LRR types of sequences,numbered from LRGA-6 to LRGA-12.Cluster analysis revealed that LRGA-1,LRGA-2,LRGA-3,LRGA-4 and LRGA-5 had high homology(59%-86%)with the known tobacco resistance gene N and flax rust resistance gene L6,while LRGA-6,LRGA-7,LRGA-8,LRGA-9,LRGA-10,LRGA-11 and LRGA-12 had high homology(51%-70%)with the known Arabidopsis resistance gene RPM1 and rice resistance gene Xa-1.In particular,LRGA-3 and LRGA-5 se-quences had extremely high similarity of amino acid sequences with various plant resistance genes to tobacco mosaic virus(TMV),suggesting that they may be anti TMV genes in Passiflora.
