Screening of antagonistic bacteria and its biocontrol effect against apple root rot
To achieve green and efficient control of apple root rot,the dilution plating method was employed to isolate bacteria from apple rhizosphere.Using the apple root rot pathogen Rosellinia necatrix as the indicator strain, antifungal bacterial strains were screened, and the indoor control experiments and strain identification of the potential strain S-25 were carried out.The re-sults showed that a total of 68 bacteria strains were isolated from the apple rhizosphere, and 17 strains were able to inhibit the growth of R. necatrix, and the strain S-25 could colonize on apple branches. The strain S-25 effectively achieved 50%inhibition rate against several apple pathogens including Fusarium solani,Botryosphaeria dothidea, and Valsa mali, with a slightly lower in-hibition rate of 48.89%for maize sheath blight pathogen,Rhizoctonia solani.Morphological characteristics and multigenetic se-quence analysis results of gyrA,16S rRNA,and rpoB suggested that strain S-25 belongs to Bacillus amyloliquefaciens. The pre-vention and control effect of S-25 on root rot disease was 61.11%and 27.78%,respectively,at one year after application,indica-ting that early application of biocontrol strain S-25 could better control the apple root rot disease.
