目的 探讨circ_0007059 对前列腺癌LNCaP细胞恶性生物学行为的影响.方法 选取 2017 年 5 月至2020 年在我院就诊的41 例前列腺癌患者的癌组织和癌旁组织标本;体外培养前列腺癌LNCaP细胞,采用质粒转染分别过表达/敲低circ_0007059 表达、抑制miR-1303 表达,并同时上调circ_0007059 和miR-1303 表达.转染后,检测circ_0007059 和miR-1303 表达、细胞增殖、迁移、侵袭,以及Ki-67、MMP-2、MMP-9 蛋白表达,并验证circ_0007059 和miR-1303 的靶向关系.结果 前列腺癌组织中circ_0007059 表达水平降低,miR-1303 表达水平升高(P<0.05).circ_0007059 过表达或抑制miR-1303 表达后,LNCaP细胞活性、迁移和侵袭能力,以及Ki-67、MMP-2、MMP-9 蛋白水平均降低.circ_0007059 靶向调控miR-1303;上调miR-1303 逆转了circ_0007059 过表达对LNCaP细胞的作用.结论 过表达circ_0007059 可抑制LNCaP细胞的恶性生物学行为,其机制可能与靶向下调miR-1303 有关.
Effect of circ_0007059 targeting miR-1303 on malignant biological behavior of LNCaP cells in prostate cancer
Objective To investigate the effect of circ_0007059 on malignant biological behavior of prostate cancer LNCaP cells.Methods The cancer tissue and paracancerous tissue specimens of 41 prostate cancer patients in our hospital from May 2017 to 2020 were selected.In vitro culture of prostate cancer LNCaP cells,plasmid transfection was used to overexpress/knockdown circ_0007059 expression,inhibit miR-1303 expression,and up-regulate circ_0007059 and miR-1303 expression,respectively.After transfection,circ_0007059 and miR-1303 expression,cell proliferation,migration,invasion,Ki-67,MMP-2,MMP-9 protein expression were detected,and the targeting relationship between circ_0007059 and miR-1303 was verified.Results The expression of circ_0007059 in prostate cancer tissue was decreased,and the expression of miR-1303 was increased(P<0.05).After circ_0007059 overexpression or inhibition of miR-1303 expression,the LNCaP cells viability,migration and invasion ability and Ki-67,MMP-2 and MMP-9 protein levels was decreased.circ_0007059 targets and regulates miR-1303;up-regulation of miR-1303 reversed the effects of circ_0007059 overexpression on LNCaP cells.Conclusions Overexpression of circ_0007059 can inhibit the malignant biological behavior of LNCaP cells,and the mechanism may be related to the targeted down-regulation of miR-1303.
