Pharmacological mechanism analysis of Shibi Decoction in treating oligoasthenozoospermia based on the spectrum-effect relationship
Objective To clarify the pharmacodynamic relationship and pharmacological substance basis of the characteristic peak component of Shibi decoction for oligoasthenozoospermia therapy.Methods The rat experiment was carried out in the study.The rats in the Shibi decoction group were orally administered with twice the equivalent human dose,while the rats in the control group was intervened with an equal volume of physiological saline.They were all continuously given medication for 7 days.The components of Shibi decoction that entered the bloodstream after oral absorption were identified by UHPLC-QE-MS technology,and the key components,key targets and enrichment pathways of Shibi decoction were obtained through network pharmacology analysis.Finally,a molecular docking analysis were performed between key blood components and core target molecules for determining spectrum-effect relationships.Results A total of 41 compounds in Shibi decoction were identified,including isosakuranetin,and 11 blood components were obtained.Shibi decoction might regulate sex hormones,anti-inflammatory and antioxidant through the estrogen signaling pathway.A visual molecular docking was formed between core targets and six components including isosakuranetin.Conclusion Shibi decoction may treat oligoasthenozoospermia by regulating lipid metabolism and endocrine disorders,and its key pharmacological ingredients are flavonoid components such as isosakuranetin.
