N+低能离子注入与亚硝酸钠复合诱变筛选子囊霉素高产菌株的研究

为了获得子囊霉素高产菌株,以链霉菌NJWGH 1322为研究对象,利用N+低能离子注入、亚硝酸钠(NaNO2)进行诱变.试验确定了最佳诱变条件:离子注入诱变剂量为200× 1014ions/cm2;NaNO2诱变时间为5min.之后通过结合2种诱变方式进行复合诱变,结果显示:复合诱变效果明显优于单因子诱变.试验最终筛选出1株产量突变提高43.1%的高产菌株,经5次传代试验表明该菌株遗传稳定性较好.
Screening of high ascomycin-producing strains by N+ ion implantation and NaNO2 complex mutation
In order to obtain high ascomycin-producing strains, the strain NJWGH1322 was used as original strain for mutation by N+ ion implantation and NaNO2. The optimal amount of ion beam was 200× 104 ions/cm2 and the mutation time of NaNO2 was 5min. The both method were combined to do complex mutation. The results showed that the effect of complex mutation was much better than that of single mutation. A high ascomycin-producing strain was obtained using complex mutation. The production of ascomycin was increased by 43.1% compared to that of the original strain. The strain showed good stability after five generations.
