富硒冬虫夏草发酵工艺优化
Optimization of fermentation conditions of selenium-rich Cordyceps sinensis

摘要
采用冬虫夏草作为富硒的载体,啤酒糟为基质,进行富硒冬虫夏草液态深层发酵试验,研究冬虫夏草对无机硒的生物转化能力.利用正交试验设计,对富硒冬虫夏草液态深层发酵条件如摇床转速、接种量、pH等进行了优化.结果表明,冬虫夏草能在含有低质量浓度亚硒酸钠(10~25mg/L)的培养基中生长,并在菌丝体内富集硒,最佳发酵工艺为摇床频率180r/min、培养10d的条件下,装液量50mL/250mL,pH值为7.0,接种量15%,温度20℃,亚硒酸钠质量浓度25 mg/L,啤酒糟质量浓度20 mg/L.在此优化的发酵条件下,富硒冬虫夏草菌丝体总富硒量为5 808.20 μg/L.
Abstract
Using Cordyceps sinensis as selenium carrier,beer distiller's grainsas substrate,the submerged fermentation experiment on selenium-rich C.sinensis was carried out to study its bioconversion capacity of inorganic selenium.The fermentation conditions of shaking speed,inoculum and pH were optimized by orthogonal experiment.Results showed that C.sinensis could grow under low concentration of sodium selenite (10-25 mg/L) and selenium was accumulated in mycelia.The optimum fermentation conditions were shaking speed 180 r/min,culture time 10 d,liquid volume 50 ml/250 ml,pH 7.0,inoculum 15%,temperature 20 ℃,sodium selenite concentration 25 mg/L,beer distiller's grains concentration 20 mg/L.Under the optimal condition,the total amount of selenium accumulation in C.sinensis mycelia was 5 808.20 μg/L.
