摘要

该研究建立了一种采用超高效液相色谱-串联质谱(UPLC-MS/MS)同时测定白酒中21种氨基酸含量的检测方法.白酒样品经过12 000 r/min离心5 min后,将上清液直接用于色谱分析,外标法定量.结果表明,在10~1 000 ng/mL质量浓度范围内,氨基酸含量线性关系良好,相关系数(R2)均>0.995,检出限和定量限分别为0.1~5.0 ng/mL和0.2~20 ng/mL,其保留时间相对标准偏差(RSD)<0.29%,峰面积RSD为1.54%~10.33%,加标回收率为80%~130%.该方法准确可靠,重复性好,可以用于白酒中21种氨基酸的含量的测定.

Abstract

A method was developed for the simultaneous determination of 21 amino acids in Baijiu using ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).After 12 000 r/min centrifugation of Baijiu samples for 5 min,the supernatant was filtered through a mem-brane for instrumental analysis and quantified by external standard method.The results showed that the standard curve was linear over the mass con-centration range of 10-1 000 ng/ml with the correlation coefficient(R2)greater than 0.995,the limits of detection were 0.1-5.0 ng/ml and the limits of quantification were 0.2-20 ng/ml,the relative standard deviation(RSD)of the retention time was less than 0.29%,the RSD of the peak area repeatability was in the range of 1.54%-10.33%,and the recovery rate of the target substance was 80%-130%.The method was accurate,reliable and reproducible,and could be used for the determination of 21 amino acids in Baijiu.
