Functional analysis of Monascus ruber M7 and MrVeAgene related mutants by light control
In order to investigate the effect of light control on the function of Monascus ruber M7 and MrVeA gene related mutant ΔMrVeA,OEMrVeA,the gene deletion strain(ΔMrVeA)and overexpression strain(OEMrVeA)were constructed by MrVeA gene knockout and overexpression techniques,respectively.Using lucifuge(dark)as control,the effects of red light and white light on the growth and development,and the production of Monascus pigment(Mps)and citrinin(CIT)of M.ruber M7,MrVeA gene mutant strains ΔMrVeA and OEMrVeA were analyzed.The results showed that the production of cleistothecium and conidium of mutant OEMrVeA increased under dark conditions,and there was no significant difference in mycelium development,colony color,biomass and pigment content compared with M.ruber M7,but the citrinin production capacity decreased by 7.03%com-pared with M.ruber M7,indicating that the overexpression of MrVeA gene could promote the sexual/asexual reproduction of strain M.ruber M7 and inhibit the citrinin formation.Red light was conducive to the growth and development of the mutant OEMrVeA,sexual/asexual reproduction and Mps generation,but was not conducive to citrinin generation of mutant strain OEMrVeA.White light was not conducive to the growth and development of mutant OEMrVeA,sexual/asexual reproduction,and Mps generation.The results could provide a reference for the industrial application of Monascus with low or even no citrinin production.
