基于斑马鱼模型的酸枣仁复合保健酒助睡眠功效和安全性研究

为研究酸枣仁复合保健酒的助睡眠功效和安全性,以受精后5 d斑马鱼为实验对象,通过水溶给予对戊四唑(PTZ)建立斑马鱼失眠模型,利用行为分析仪测定模型斑马鱼和正常斑马鱼的觉醒活动量和觉醒总时间,评价酸枣仁复合保健酒改善睡眠功效。以受精后2d斑马鱼为实验对象,通过斑马鱼组织器官毒性发生率评价酸枣仁复合保健酒安全性。结果表明,酸枣仁复合保健酒具有改善模型斑马鱼和正常斑马鱼睡眠的功效,具体表现为:显著降低模型斑马鱼的觉醒活动量和觉醒总时间(P<0。05),降低了正常斑马鱼的觉醒活动量和觉醒总时间。斑马鱼急性毒性实验结果显示,酸枣仁复合保健酒在2。83 µL/mL给予量下对斑马鱼是安全的,超过8。48 μL/mL给予量对斑马鱼产生毒性作用。综上,酸枣仁复合保健酒对斑马鱼具有助睡眠功效。
Sleep aid function and safety of complex healthy liquor of spina date seed based on zebrafish model
In order to study the sleep aid effect and safety of complex healthy liquor of spina date seed,using the zebrafish 5 d after fertilization as the experimental subjects,a zebrafish insomnia model was established by administering p-pentatetrazole(PTZ)by water solution.The waking activity and total waking time of model zebrafish and normal zebrafish were measured by behavior analyzer,and the sleep improving effect of the liquor was eval-uated.Taking zebrafish 2 d after fertilization as the experiment object,the safety of the liquor was evaluated by the incidence of tissue and organ toxi-city of zebrafish.The results showed that the liquor had the effect of improving the sleep of model zebrafish and normal zebrafish,which showed as follows:significantly decreased the waking activity and the total waking time of model zebrafish(P<0.05),and decreased the waking activity and the total waking time of normal zebrafish.The results of acute toxicity test of zebrafish showed that the liquor was safe to zebrafish at administration of 2.83 μl/ml,and toxic to zebrafish when the administration exceeded 8.48 μl/ml.To sum up,the complex healthy liquor of spina date seed had the ef-fect of sleep aid function on zebrafish.
