黄精多糖酶辅助法提取工艺优化及活性分析研究

该试验以黄精为原材料,采用酶辅助法提取黄精多糖,通过单因素及正交试验优化提取工艺,通过透析获得不同截留分子质量黄精多糖片段进行吸湿保湿性试验,并考察最佳保湿片段的抗氧化能力及结构表征分析。结果表明,在复合酶添加量6%、酶解时间90min、酶解温度50℃、料液比1∶30(g∶mL)的条件下,黄精多糖得率达到24。18%。当黄精多糖分子质量>8 000Da时,其保湿性能最佳;当黄精多糖最佳保湿片段质量浓度为6mg/mL时,对1,1-二苯基-2-三硝基苯肼(DPPH)、2,2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)、羟基自由基的清除率分别为50。54%、48。65%、26。41%,具有一定的抗氧化活性;同时发现,最佳保湿片段表面基本平滑,呈不规则的薄片且分子间存在-OH、C-H、C=O等官能团。
Optimization of enzyme assisted extraction process and activity analysis of Polygonatum sibiricum polysaccharide
Using Polygonatum sibiricum as raw material,P.sibiricum polysaccharide was extracted by enzyme assisted method and the extraction pro-cess was optimized by single factor and orthogonal tests.The moisture absorption and moisture retention tests were carried out with P.sibiricum polysaccharide fragments of different molecular weight obtained by dialysis,and the antioxidant capacity and structural characterization of the optimal moisturizing fragments were investigated.The results showed that the polysaccharide yield reached 24.18%under the conditions of compound enzyme addition 6%,enzymolysis time 90 min,enzymolysis temperature 50 ℃,solid and liquid ratio 1∶30(g∶ml).When the mass concentration of the optimal moisturing fragment of P.sibiricum was more than 8 000 Da,its moisturing property was optimal.When the mass concentration of the optimal mois-turing fragment of P.sibiricum polysaccharide was 6 mg/ml,the scavenging rates of DPPH,ABTS+and hydroxyl radicals were 50.54%,48.65%and 26.41%,respectively,which showed certain antioxidant activities.At the same time,it was found that the surface of moisturing fragment was basically smooth,with irregular flakes and intermolecular functional groups such as-OH,C-H and C=O.
