Screening and identification of β-glucosidase-producing strain and enzymatic properties of crude enzyme
β-glucosidase-producing strains were selected from the soil of decayed trees in primitive forests,and the β-glucosidase production ability was measured.The morphological characteristics,molecular biological identification and enzymatic properties of the high-yield strains were studied.The results showed that 13 β-glucosidase-producing strains were screened,a high-yield glucosidase strain PX14A was obtained through re-screening,and the β-glucosidase activity was 4.9 U/ml.The strain PX14A was identified as Bacillus subtilis.The results of enzymatic properties research showed that the β-glucosidase of the strain PX14A crude solution was stable below 65 ℃,the optimal reaction temperature was 60 ℃,the enzyme was stable at pH 4.0-7.5,and the optimal reaction pH value was 6.5.Mg2+and Fe2+with a concentration of 1 mmol/L had a good activating effect on enzyme ac-tivity.The results indicated that the strain PX14A had good application prospects.
