摘要
该研究从原始森林腐木的土壤中取样,筛选产β-葡萄糖苷酶菌株,对其产β-葡萄糖苷酶能力进行测定,并对高产菌株进行形态特征、分子生物学鉴定及酶学性质研究.结果表明,筛选得到13株产β-葡萄糖苷酶菌株,复筛得到1株高产β-葡萄糖苷酶的菌株PX14A,其β-葡萄糖苷酶酶活为4.9 U/mL.菌株PX14A被鉴定为枯草芽孢杆菌(Bacillus subtilis).酶学性质研究结果表明,菌株PX14A粗酶液的β-葡萄糖苷酶在65 ℃以下稳定,最适反应温度为60 ℃,在pH值4.0~7.5较稳定,最适反应pH值为6.5,浓度为1 mmol/L的Mg2+和Fe2+对酶活性有较好的激活作用.结果显示该菌株具有良好应用前景.
Abstract
β-glucosidase-producing strains were selected from the soil of decayed trees in primitive forests,and the β-glucosidase production ability was measured.The morphological characteristics,molecular biological identification and enzymatic properties of the high-yield strains were studied.The results showed that 13 β-glucosidase-producing strains were screened,a high-yield glucosidase strain PX14A was obtained through re-screening,and the β-glucosidase activity was 4.9 U/ml.The strain PX14A was identified as Bacillus subtilis.The results of enzymatic properties research showed that the β-glucosidase of the strain PX14A crude solution was stable below 65 ℃,the optimal reaction temperature was 60 ℃,the enzyme was stable at pH 4.0-7.5,and the optimal reaction pH value was 6.5.Mg2+and Fe2+with a concentration of 1 mmol/L had a good activating effect on enzyme ac-tivity.The results indicated that the strain PX14A had good application prospects.
