Optimization of high-density culture conditions for Weizmannia coagulans
In order to meet the increasing market demand for Weizmannia coagulans and obtain a method for high-density culture of W.coagulans,using W.coagulans D1 as the experimental object,based on MRS medium,the high-density culture conditions of W.coagulans were optimized through single factor experiments and response surface experiments,and 10 L amplification culture was carried out under the optimal conditions.The results showed that the optimal culture conditions were culture temperature 50 ℃,initial pH 8.0,time 16 h.The optimal medium formulation was glucose 24 g/L,yeast extract powder-tryptone(1∶1)31 g/L,magnesium sulfate heptahydrate 0.29 g/L.After optimization,the OD620nm value of the strain reached 2.15,which was 68%higher than that before optimization,and the lag phase was shorter.Based on the above experimental results,the OD620nm reached 7.90 by setting the 10 L fermenter with stirring speed of 200 r/min and aeration culture.The results provided a basis for the industrial production of W.co-agulans and lay a foundation for the subsequent production of bacterial powder.
