Study on the detection and stability of 8 effective components in compound health wine
A method for the determination of 8 effective components(5-hydroxymethylfurfural,adenosine,cordycepin,N6-(2-hydroxyethyl)adenosine,picrocrocin,rutin,crocin Ⅰ and crocin Ⅱ in compound health wine was established by HPLC,and the stability of 8 effective components was analyzed.The results showed that the optimal chromatographic conditions were as follows:Agilent SB-Aq C18 column(4.6 mm×250 mm,5 μm),acetonitrile-0.1%acetic acid aqueous as mobile phase,flow rate 1.0 ml/min,detection wavelength 260 nm/440 nm,column temperature 30 ℃,and injection volume 10 μl.Under the analytical conditions,the 8 effective ingredients showed good linear relationships in their respective mass concentration ranges(R2≥0.999 9).The limits of detection(LOD)were 15.5-146.1 μg/L,and the limits of quantitation(LOQ)were 45.5-429.7 μg/L.The relative standard de-viations(RSD)of precision,stability and repeatability tests were all lower than 2.0%.The average spiked recovery rates were 96.15%-99.23%.The stability tests results of samples showed that the adenosine,cordycepin,N6-(2-hydroxyethyl)adenosine and picrocrocin were stable under different storage conditions for 90 d,the 5-hydroxymethylfurfural was sensitive to temperature,and the rutin,crocin Ⅰ,and crocin Ⅱ were extremely sensitive to light and heat.
