Application study of DNA ploidy analysis combined with HPV DNA detection in screening of cervical lesions
Objective To explore the application value of DNA ploidy analysis combined with HPV DNA detection in screening cervical lesions. Methods Patients who underwent cervical lesion screening at Changning District Maternal and Child Health Hospital in Shanghai from January to June 2021 were selected.DNA ploidy analysis was performed first, and HPV DNA testing was performed for those with abnormal results. Finally, cervical biopsy samples were taken for patients who were both positive, and were compared with the pathological results of cervical biopsy. Results A total of 9126 cervical samples underwent DNA ploidy examination, with 680 cases showing abnormal results. After HPV DNA combined detection,503 cases underwent hysteroscopic cervical biopsy and were sent for pathological examination. The results showed that the proportions of cervical cancer,high-grade squamous intraepithelial lesions(HSIL),low-grade squamous intraepithelial lesions(LSIL),and cervicitis were 1.6%(8/503),11.5%(58/503),41.4%(208/503),and 45.5%(229/503),respectively.The positive rate of LSIL and above in groups with DNA ploidy abnormalities≥3 was 39.1%(107/274).After combined HPV DNA examination, the positive rate of LSIL and above was increased to 99.6%(275/276).The difference in positive rates between the two groups was statistically significant(P<0.05),while the dif-ference in negative rates was not statistically significant(P>0.05).Conclusion The combination of DNA ploidy analy-sis with HPV DNA detection can improve the positive rate of cervical lesion screening while maintaining the same nega-tive rate, which has clinical significance.
