Screening of Stropharia rugosoannulata Strains with High Yield Polysaccharide by ARTP Mutagenesis and Optimization of Mycelial Culture Conditions
In order to obtain excellent strains of Stropharia rugosoannulata,atmospheric and room temperature plasma mutagen-esis was used to treat S.rugosoannulata spores.By exploring the growth vigor,antagonistic effects,and polysaccharide content of the mutated mycelium,the mutant strains with fast mycelial growth and high polysaccharide content were screened,and the mycelial polysaccharide structural feature and infrared spectroscopy,and mycelium liquid culture conditions of the mutant strain were investigated.The results showed that among the 42 screened mutagenic strains,5 mutant strains with good growth poten-tial,genetic stability and antagonism to the original strain were obtained.Among them,the mycelium of Dl and D4 strains had higher biomass and polysaccharide content,the mycelium polysaccharide particles of D1 strain were uniform and regular,and the particle size was about 0.05 pm,which had the infrared spectrum characteristics of general polysaccharides.The suitable liquid culture conditions for D1 strain mycelium are spore inoculation rate of 8%,rotation speed of 115 r·min-1,culture temper-ature of 24 ℃,and culture time of 4 days,the biomass of mycelium under these conditions was 56.82 mg.
