1株野生血红栓菌的分离鉴定及生物学特性分析

从曲阜孔林采集到1株野生担子菌子实体,通过组织分离获得其菌丝,编号为YEF202102.进行了形态学特征分析、ITS测序及系统发育树构建,发现菌株YEF202102的形态特征与血红栓菌相符,与血红栓菌ITS序列相似度达到99%,并与其在系统发育树上聚在一支,自聚值为100%,鉴定为血红栓菌(Trametes Sanguinea).为开发利用该野生资源,通过单因素试验和正交试验探究了菌株YEF202102在固体培养条件下各因素对菌丝生长的影响.结果显示,最适合菌株YEF202102菌丝生长的条件为麦芽糖20 g·L-1、酵母粉2 g·L-1,pH 5.0,温度 25℃.
Pure culture strain(numbered as YEF202102)was from the wild fruit bodies of basidiomycete from the Confucian Cemetery,Qufu City.Through morphological characteristics analysis,ITS sequencing and phylogenetic tree construction,the re-sults showed that the morphological characteristics of this strain were consistent with that of Trametes.sanguinea,and the simi-larity of ITS sequence with T.sanguinea reached 99%,and the strain was clustered in the phylogenetic tree with a self-aggrega-tion value of 100%.It was therefore identified as T.Sanguinea.The biological characteristics of this fungus were studied.The results showed that,the best condition for mycelia growth of strain YEF202102 was fructose 20 g·L-1,yeast paste 2 g·L-1,pH 5.0 and temperature 25℃.
