六妹羊肚菌原生质体再生菌株的制备与比较

摘要
试验对六妹羊肚菌原生质体的制备和再生条件进行了研究,并对再生菌株开展菌丝体生长速度测定和栽培试验.结果表明,原生质体形成的最适条件为选择培养3 d的菌丝体,以0.6 mol·L-1的甘露醇作为稳渗剂,添加1.5%的溶壁酶B,30℃酶解3h.该条件下原生质体产量高达8×104个/mL.再生菌株在菌丝形态、细胞结构、菌丝生长速度方面都和原始菌株有很大差异,其中Z2和Z4菌株的菌丝在不同培养基上的生长速度均较快,且在栽培时产量分别达到427.3g·m-2和380.3g·m-2,可作为种质资源开展进一步试验.
Abstract
The protoplast formation and regeneration conditions of Morchella sextelata were studied,the mycelium growth rates of the regenerated strains was measured and the cultivation experiment was carried out.The results showed that the optimum condition for protoplast formation was to select the mycelium cultured for 3 d,use 0.6 mol·L-1 mannitol as osmotic stabilizer,add 1.5%lyticase B,and the enzymolysis was conducted at 30 ℃ for 3 h.Under this condition,the protoplast yield reached 8x 104 per mL.The regenerated strain had great differents from the original strain in mycelia morphology,cell structure and mycelial growth rate.The mycelium growth rate of regenerated strain Z2,Z4 groups in different media was faster,and during cultivation the yield reached 427.3 g·m-2 and 380.3 g·m-2 respectively,which could be used as germplasm resources for further investigation.
