Isolation,Identification of Inocutis levis CM01 and Antitumor Activity Analysis of Its Fermentation Broth
Pure culture mycelium was obtained by tissue isolation from a wild strain Inocutis levis CM01,and the strain was i-dentified based on morphology and ITS sequence analysis.The antitumor compounds in the fermentation broth of this strain were isolated and purified by high performance liquid chromatography(HPLC),and the inhibitory activities against HeLa tumor cells were analyzed by MTT and flow cytometry.The results showed that the isolated strain CM01 had the highest homology similarity with Inocutis levis(95%).Combined with morphological analysis,CM01 was identified as a new strain of Inocutis levis.The fermentation broth of this strain can inhibit HeLa,MCF-7 and B16 tumor cells,and the active component TMA con-taining cyclic peptide compounds can be isolated from the fermentation broth.The half inhibitory rate of TMA on HeLa tumor cells is 12 µg·min-1,and the replication cycle of HeLa cells can be arrested in the S phase.
