黑木耳遗传多样性分析及高产多糖菌株筛选

利用体细胞不亲和性分析、ISSR分子标记对东北三省地区主栽及收集的 45 份黑木耳种质资源进行遗传多样性分析,并通过液体深层发酵技术筛选高产多糖菌株.结果表明,其中 9 个菌株拮抗反应不明显,亲缘关系较近;其他 36个菌株的遗传相似系数变化范围为 0.44~0.96,遗传背景丰富.当遗传相似系数为 0.6 时,供试菌株聚类为 9个类群:黑山、黑丰、黑丰 2、神 8-7、黑威伴金、黑威 15、黑保 1 号、2016-A、2016-B、2016-C、2016-E、2019-013、2019-192、新科 10 号、黑厚圆、华腾 2 号、华腾 4 号、春宝为第 1 类群;木 29、长白7、新科 1 号、919 为第 2 类群;高优王、1022、2019-003、918 为第 3 类群;蕈丰黑、延丰 13、半筋S1908、888、丰产王为第 4类群;靖发 13、89-1、丰收 2号、2019-199、海元的遗传相似系数小于 0.6,亲缘关系较远,单个菌株自成一类,分别为第 5~9类群.经筛选,得到 1株多糖含量高且丰产的黑木耳菌株黑丰 2.
Genetic Diversity Analysis and Strains Screening of Auricularia auricula with High Polysaccharide Yield
The genetic diversity of 45 Auricularia auricula germplasm resources collected from three northeastern provinces was analyzed by somatic incompatibility analysis and ISSR molecular marker,then high yield polysaccharide strains of which were screened by the liquid deep fermentation technology.The results showed that 9 strains had no obvious antagonistic reaction and were closely related to each other.The genetic similarity coefficient of the other 36 strains ranged from 0.44 to 0.96,and the genetic background was rich.When the genetic similarity coefficient was 0.6,the tested strains were clustered into nine groups:Heishan,Heifeng,Heifeng 2,Shen 8-7,Heiwei Banjin,Heiwei-15,Heibao No.1,2016-A,2016-B,2016-C,2016-E,2019-013,2019-192,Xinke No.10,Heihouyuan,Huateng No.2,Huateng No.4 and Chunbao were the first group;Mu 29,Changbai 7,Xinke No.1 and 919 were the second group;Gaoyouwang,1022,2019-003 and 918 were the third group;Xunfenghei,Yanfeng-13,Banjin S1908,888 and Fengchanwang were the fourth group;The genetic similarity coefficient of Jingfa 13,89-1,Fengshou No.2,2019-199 and Haiyuan was less than 0.6,indicating that the genetic relationship was distant,and the single strain as sui generis,which were the fifth to the ninth groups respectively.From these strains,A.auricula strain Heifeng 2 with high polysaccharide content and high yield was screened out.
