摘要
目的 检测形觉剥夺性近视形成过程中视网膜代谢变化.方法 14天豚鼠被随机分为形觉剥夺模型组(48只),右眼单眼覆盖剥夺面罩(FD眼),左眼不做覆盖(F眼);及对照组F眼(20只).分别在造模3d和14 d时取两组动物右眼视网膜进行气相一时间飞行质谱检测.结果 (1)接受形觉剥夺3d和14 d后,FD眼屈光度比对照眼下降,玻璃体腔深度(VCD)及眼轴(AL)有增长趋势.(2)与对照组相比,共有11种代谢物在造模期间发生变化;其中3种代谢物(谷氨酸、葡萄糖、酪氨酸)仅在造模3d时发生变化,4种(异亮氨酸、苏氨酸、丙氨酸、苹果酸)在造模3d和造模14 d均发生变化,4种(十八碳烯酸、十八烷酸、花生四烯酸、十六烷酸)仅在造模14 d时发生变化.结论 葡萄糖堆积和不饱和脂肪酸合成水平可能与近视的形成和发展有密切关系.
Abstract
Objective To evaluate changes in retinal metabolic profiles during form deprivation.Methods Animals were randomly divided into control group (20 animals) and form deprivation group (48 animals).Right eyes of animals in form deprivation group were covered by a mask (FD eye),while the left eyes (F eye) were uncovered.Gas chromatography time-of-flight mass spectrometry was applied to compare guinea pig retinal metabolite 1evels in form deprivation (FD) eyes at 3 days and 2 weeks post FD with normal control (NC) eyes.Results (1) After 3 days and 14 days of FD,the refraction of FD eyes were decreased and the vitreous cavity depth and axes length of FD eyes were increased,compared with normal control animals.(2) Compared with the ipsilateral eyes of normal control group,11 metabolites were changed,among which 3 metabolites (glutamate,glucose,tyrosine) were changed only in 3-day-FD group,4 metabolites (isoleucine,threonine,alanine,malic acid) were changed both in 3-day and 14-day FD group,and 4 metabolites (octadecenoic acid,octadecanoic acid,arachidonic acid and hexadecenoic acid) were changed only in 14-day FD group.Conclusions Myopia progression is associated with an inverse relationship between increases in glucose accumulation and lipid level decreases in form-deprived guinea pig eyes.
