本文旨在从一种未应用于甜菜的CEAP引物中筛选出多态性高、稳定性强、重复性好的核心引物,并找到适合检测CEAP引物多态性的方法.本研究选取了 4 个与植物生长发育、信号转导和胁迫响应密切相关的顺式元件GATAA、TGAC、AAAG和ACGTG为核心的CEAP分子标记,通过对部分甜菜品种的基因组进行扩增,并利用适宜的检测方法,对83 条CEAP引物多态性进行筛选.结果表明,最优的检测方法为2%浓度的琼脂糖凝胶;从83 条CEAP引物中筛选出17 条扩增产物条带清晰稳定、多态性高的引物,多态性信息含量范围为0.2608~0.8446,平均值为0.6649;表明CEAP引物具有较高的多态性,筛选的引物可以用于今后甜菜品种指纹图谱的构建以及遗传多样性分析.
Screening of Sugar Beet CEAP Core Primers
In this paper,we aimed to select core primers with high polymorphism,strong stability and repetition from a CEAP primer not applied to sugar beet,and to find a suitable method for detecting CEAP primer polymor-phism.In this study,four CEAP molecular markers of cis elements,GATAA,TGAC,AAAG and ACGTG,closely related to plant growth development,signal transduction and stress response,were selected to amplify the genome of some beet varieties and screen 83 CEAP primer polymorphisms by using appropriate detection meth-ods.The results show that the optimal detection method is 2%agarose gel;17 primers from 83 CEAP primers,the polymorphism information content ranges from 0.2608 to 0-0.8446,the average is 0.6649;indicating that CEAP primers have high polymorphism,which can be used for the construction of beet fingerprint and genetic di-versity analysis.
