Optimization of Ultrasonic-Assisted Extraction Process of Flavonoids from Cantonese Tangerine Peel and Study on Their Antioxidant Activity
With Cantonese tangerine peel produced in Xinhui,Guangdong as the raw material,the ultrasonic-assisted extraction process of flavonoids from Cantonese tangerine peel is optimized by orthogonal test method,and the antioxidant activity is determined.With rutin as the reference,a quantitative analysis method for the determination of flavonoid content in Cantonese tangerine peel is established by ultraviolet and visible spectrophotometry.Based on single factor test,orthogonal test method is used to analyze the interaction of ethanol concentration,ultrasonic time,ultrasonic power and solid-liquid ratio,so as to obtain the best conditions and verify them.Finally,the antioxidant capacity of flavonoids from Cantonese tangerine peel is studied by the method of scavenging DPPH free radicals and hydroxyl free radicals.It is concluded that when the ethanol concentration is 60%,the ultrasonic time is 45 min,the ultrasonic power is 300 W and the solid-liquid ratio is 1∶50,the extraction rate of flavonoids from Cantonese tangerine peel could reach 1.86%.With the increase of flavonoid concentration,the scavenging rates of DPPH and hydroxyl free radicals are higher,indicating that the flavonoids from Cantonese tangerine peel have higher antioxidant capacity and they are a strong antioxidant.
