山楂不同提取物成分分析及其生物活性研究

山楂富含果糖、葡萄糖、脂肪类物质、蛋白质、粗纤维和胡萝卜素等,具有抗炎、降血糖和降血压等生物活性功能,被广泛应用于食品添加剂、调味品和保健品等方面.山楂中活性成分提取方式多样,不同提取方式获得的化合物成分和含量存在差异.目前,溶剂提取法是最常见的提取方式,该方法技术要求低、操作简单,但是不同溶剂提取获得的化合物成分和含量存在明显差异.该试验采用高效液相色谱法测定6种不同溶剂(甲醇、乙醇、丙酮、乙酸乙酯、氯仿和石油醚)的山楂提取物中多酚、黄酮等活性成分含量,结果表明,山楂甲醇提取物中多酚和黄酮含量最高,山楂石油醚提取物中多酚和黄酮含量最低;6种不同溶剂的山楂提取物中鉴定出10种活性成分,分别为儿茶素、绿原酸、原花青素B2、表儿茶素、槲皮素、山楂酸、金丝桃苷、异槲皮苷、咖啡酸、木犀草素葡萄糖甙.通过对不同溶剂提取的山楂化合物的抗氧化性和还原性进行分析,结果表明,随着山楂提取物浓度的增加,抗氧化性和还原性也增加,山楂甲醇提取物的抗氧化性和还原性相较于其他溶剂的山楂提取物更强.
Analysis of Components and Biological Activity of Different Extracts from Hawthorn
Hawthorn is rich in fructose,glucose,fatty substances,protein,crude fiber,carotene and so on.It has biological activities such as anti-inflammatory,regulating blood sugar level and reducing blood pressure,and is widely used in food additives,condiments,health care products and so on.There are various methods for extracting active components from hawthorn,and the composition and content of compounds obtained by different extraction methods are different.At present,solvent extraction method is the most common extraction method.This method requires low technical requirements and is easy to operate,but there are obvious differences in the composition and content of compounds obtained by different solvents.In this experiment,the content of polyphenols,flavonoids and other active components in hawthorn extracts of six different solvents(methanol,ethanol,acetone,ethyl acetate,chloroform and petroleum ether)is determined by high performance liquid chromatography.The results show that the content of polyphenols and flavonoids in hawthorn methanol extract is the highest,and the content of polyphenols and flavonoids in hawthorn petroleum ether extract is the lowest;there are 10 active components identified in hawthorn extracts of six different solvents,which are catechin,chlorogenic acid,proanthocyanidin B2,epicatechin,quercetin,maslinic acid,hyperoside,isoquercitrin,caffeic acid and luteolin glucoside.The antioxidant and reducing properties of hawthorn compounds extracted by different solvents are analyzed.The results show that with the increase of the concentration of hawthorn extracts,the antioxidant and reducing properties increase,and the antioxidant and reducing properties of hawthorn methanol extract are stronger than those of other solvents.
