石榴皮提取物指纹图谱的建立及指标成分含量测定

为建立石榴皮提取物的指纹图谱及指标成分含量测定的高效液相色谱方法,为石榴皮提取物质量控制提供参考,文章采用高效液相色谱法建立石榴皮提取物指纹图谱,并通过相似度评价结合化学计量法分析10个批次石榴皮提取物的质量差异,同时以安石榴苷和鞣花酸作为指标成分建立含量测定方法.结果表明,建立10个批次石榴皮提取物指纹图谱,共28个共有峰,其中8号峰为α-安石榴苷,13号峰为β-安石榴苷,28号峰为鞣花酸,S2、S4、S10 3个批次的石榴皮提取物的相似度<0.9,聚类分析将10个批次石榴皮提取物分为5类,通过VIP值筛选确定安石榴苷和鞣花酸为主要差异性指标成分,测得安石榴苷和鞣花酸含量分别为28.80~288.90 mg/g和6.12~17.73 mg/g.该研究建立的指纹图谱及含量测定方法可为石榴皮提取物质量控制与评价提供参考.
Establishment of Fingerprint of Pomegranate Peel Extracts and Determination of Content of Index Components
To establish a high performance liquid chromatography for the fingerprint of pomegranate peel extracts and determination of the content of index components,and to provide references for quality control of pomegranate peel extracts,in this paper,the fingerprint of pomegranate peel extracts is established by high performance liquid chromatography,and the quality difference of ten batches of pomegranate peel extracts is analyzed by similarity evaluation combined with chemometrics method.At the same time,with punicalagin and gallogen as the index components,the content determination method is established.The results show that a total of 28 common peaks are identified in the established fingerprints of ten batches of pomegranate peel extracts,among which,No.8 peak is α-punicalagin,No.13 peak is β-punicalagin,and No.28 peak is gallogen.The similarity of pomegranate peel extracts from batches S2,S4 and S10 is<0.9.The ten batches of pomegranate peel extracts are divided into five categories by cluster analysis,and through VIP value screening,punicalagin and gallogen are identified as the main differential index components.The content of punicalagin and gallogen is measured to be 28.80~288.90 mg/g and 6.12~17.73 mg/g respectively.The fingerprint and content determination method established in the research can provide references for quality control and evaluation of pomegranate peel extracts.
