目的:探讨刺梨多糖提取和纯化的方法.方法:采用热水提取醇沉法从刺梨果实中提取多糖,在单因素实验的基础上,以响应面实验对刺梨多糖的提取工艺进行优化.采用Cellulose DEAE-52和Sephadex G-100柱对刺 梨粗多糖进行分离纯化,Sevage法脱蛋白,D-101吸附树脂脱色素,FT-IR、HPGPC和HPLC测定分子量和单糖组成.结果:刺梨多糖是一种以Glc为主要成分的均一性酸性多糖.
Extraction,Purification and Structure Analysis of Polysaccharides from Rosa roxburghii Tratt.Produced in Yunnan Province
Objective:To study the extraction and purification methods of polysaccharides from Rosa roxburghii Tratt..Methods:Polysaccharides are extracted from Rosa roxburghii Tratt.fruit by hot water extraction and alcohol precipitation method.On the basis of single factor experiment,the extraction process is optimized by response surface experiment.The crude polysaccharides are separated and purified by Cellulose DEAE-52 and Sephadex G-100 columns,deproteinized by Sevage method,decolorated by D-101 adsorptivon resin,and the molcecular weight and the composition of monosaccharides are determined by FT-IR,HPGPC and HPLC.Results:Rosa roxburghii Tratt.polysaccharide is a kind of homogeneous acid polysaccharide with Glc as the main component.
