Study on Preparation and Properties of Zein-Dextran-Lutein Microcapsules
Lutein is sensitive to light and heat,which leads to its poor stability. In order to solve this problem,lutein is microencapsulated. In this paper,with lutein as the core material and zein and dextran as the wall materials,lutein is embedded by antisolvent precipitation method.The preparation process of lutein microcapsules is optimized by response surface method on the basis of single factor test,and they are characterized and their storage stability and antioxidant activity are characterized. The results show that the optimal preparation conditions of lutein microcapsules are ethanol concentration of 84%,the volume ratio of solvent to antisolvent of 1∶2,reaction temperature of 39 ℃,zein concentration of 4.0 mg/mL and dextran concentration of 1.0 mg/mL. At this time,the embedding rate is 90.53%,the drug loading rate is 8.23%. After embedding,the saturation dissolution rate of lutein increases by 4.84 times. After six weeks of storage,the retention rate of lutein microcapsules is 35.20% higher than that of lutein under light conditions,23.11% higher than that of lutein at 25 ℃,and 16.31% higher than that of lutein at 4 ℃. At-20 ℃,the retention rate of lutein microcapsules only decreases by 1.91%. The antioxidant activity of lutein microcapsules against DPPH and ABTS+free radicals is higher than that of unembedded lutein. The research results can provide theoretical references for the application of lutein microcapsules as new additives in the industrial field.
